GalNAc Oligonucleotides(GalNAc-siRNA,GalNAc-ASO,GalNAc-mircoRNA)
GalNAc (N-acetylgalactosamine) coupling modification is currently the most commonly used small nucleic acid drug delivery system for targeting the liver. Currently, 5 out of 6 siRNA drugs on the market worldwide use GalNAc covalent linkage delivery system. GalNAc is a high affinity targeting ligand for sialic acid receptor (ASGPR). GalNAc binds to ASGPR on the surface of liver cells and enters the cell to form an endosome, thereby introducing a sufficient amount of siRNA into the cell and inducing selective RNA silencing response (the mechanism of action is shown in the figure below).
